Cardiovascular disorders related to HMGB1 over-expression
Bio3 Research has entered into a strategic alliance with Syngen Inc. (CA/USA) and Protelix Inc. (CA/USA) for the development, under their technological platforms, of SiRNA HMGB1 specific and Mimic Abs anti-HMGB1 in cardiovascular field (e.g. Restenosis and Atherosclerotic Plaque progression inhibition).
SiRNA (Small Interfering RNA) HMGB1 specific have been designed to inhibit the expression of the protein HMGB1 at cell level without transfection. In vitro tests (also with optimized/purified SiRNA) have been successfully performed to verify SiRNA specificity against HMGB1.A lead has been selected to perform Proof of Concept in animals in 2009
Mimic-Abs HMGB1-specific have been developed by utilizing ProtElix pioneering technology. This approach may accelerate the development of new therapeutic tools with better tissue penetration, lower production costs and longer shelf-life. In vivo tests on Mimic Abs to verify their activity against HMGB1 will be available by end 2008 . Proof of concept is performed at BioQuant San Diego CA.
Bio3 Research has filed, in co-ownership with Prof. Marco Bianchi, Group Leader, Chromatin Dynamics Unit, San Raffaele Scientific Institute (Milan - Italy), two patent applications in the cardiovascular field comprising restenosis and atherosclerosis . Under these patents, Bio3 has the right to develop monoclonal antibodies (MABs) small molecules, HMGB1 fragments and antagonists that inhibit HMGB1 to prevent smooth muscle cell migration and endothelial cell proliferation. Patents have already been granted in Italy and Australia; nationalization phase in progress in USA, Canada, Mexico, the EU, Japan, China.
